Karel af Lothringen blev født i 1712 i Lunéville, nær Nancy. Han var bror til Frans af Lothringen og dermed svoger til Maria Theresia af Østrig. Han giftede sig også med hendes søster Maria Anne af Østrig.
Han blev i 1741 stedfortræder for de Sydlige Nederlande. Den 26. marts 1744 flyttede han ind i Nassau-paladset i Bruxelles og på slotet i Tervuren.
Af Maria Theresia af Østrig blev han leder af den Østrigske hær, men han måtte lade sig besejre i Leuthen i 1757 af en endnu mere genial feltherre Frederik den Store, konge af Preussen. Dette kostede ham hans job som øverste chef, hvorefter han trak sig tilbage til Bruxelles, hvor han optrådte som beskytter af kunsten. Han var meget elsket af folket, og allerede mens han levede, fik han to statuer: den første på facaden af Brouwershus på den Store Marked, og den anden på Kongens Plads.
Ifølge Michael Baigent var han lederen af Prieuré de Sion.
