Da kejsar Karl V undertrykte den Ghentiske opstand i år 1540, tvang han indbyggerne i Gent til at nedrive store dele af Sankt Baafs Kloster og Sankt Baafsvillaget for at anlægge Castrum Novum, det senere Spanske Slot. Klostertårnet blev revet ned, og Karl lagde selv den første sten til slottet den 12. maj 1540. Tusindvis af arbejdere gik i gang. Det enorme slot stod færdigt i 1545 og husede omkring 2500 mand.
