Großharthau er en by og den tilhørende kommune i sydvestlige del af den saksiske Landkreis Bautzen, beliggende ved Bundesstraße 6 halvvejs mellem Dresden og Bautzen og cirka 25 km fra begge byer. Kommunen danner sammen med Frankenthal et administrationsfællesskab kaldet Großharthau.
Byen Großharthau ligger omtrent seks kilometer vest-sydvest for Bischofswerda på begge sider af Bundesstraße 6, hvor den større del af bebyggelsen findes nord for denne vej. Mellem Seeligstadt og Großharthau krydser jernbanelinjen Dresden–Görlitz den nordvest for Großharthau beliggende Massenei, inden den når stationen Großharthau i den sydvestlige del af byen.
