Premnitz ligger i lavlandet i det vestlige Havelland ca. 65 kilometer vest for Berlin. Nærliggende større byer er Rathenow, Brandenburg an der Havel, Potsdam og Stendal. Premnitz ligger i Naturparken Westhavelland. Gennem byområdet strækker naturbeskyttelsesområdet Under Havel Syd sig. Derudover er der yderligere beskyttelsesområder.
Stedet blev første gang nævnt i skriftlige kilder som "Prebenitz" i 1375. Med industrialiseringen, der begyndte i slutningen af det 19. århundrede, ændrede det indtil da landsbymæssige bybillede sig.
