Vaison-la-Romaine er en fransk kommune i departementet Vaucluse i regionen Provence-Alpes-Côte d'Azur. Den er hovedby i den navngivne kanton Vaison-la-Romaine i arrondissementet Carpentras. Vaison-la-Romaine har 5.908 indbyggere og ligger ved bredden af Ouvèze og ved en krydsning af antikke romerske veje. I 1924 tilføjede kommunen sig attributten la Romaine. Dens romerske navn er Vasio Vocontiorum. Den var midlertidigt bispesæde efter det 4. århundrede. Kommunens område tilhører den regionale naturpark Mont-Ventoux.
