Die erste Erwähnung der Beginen in Mechelen stammt aus dem Jahr 1209. Nach vielen Jahren der Ansiedlung außerhalb der Stadtmauern kehrten sie 1578 in die Innenstadt zurück.
1629 wurde die zuvor errichtete Kapelle durch eine Kirche ersetzt. Pieter Huyssens und Jacques Francart zeichneten ein typisches Beispiel barocker Architektur für die Beginenhofkirche (1629-1647) . Das Design verband italienische Vorbilder mit einem regionalen Design, das nicht vollständig mit der Vergangenheit brach. Nach 1640 könnte Lucas Faydherbe, ein Schüler von Rubens, für einen Teil der Innenausstattung verantwortlich gewesen sein. Italienische Einflüsse sind durch einen Rhythmus von gewölbten Arkaden erkennbar, die von korinthischen Säulen und einem imposanten Gesims getragen werden. Die Kreuzgewölbe beziehen sich auf den gotischen Stil.
