Plymouth ist heute Standort der königlich-britischen Marinewerft. Der Marinestützpunkt Devonport im Westen der Stadt ist der größte Marinehafen Westeuropas und wichtigster Arbeitgeber in Plymouth.
Die Stadt liegt am nördlichen Ende des Plymouth Sound. Die westliche Stadtgrenze bilden der Hamoaze und der Tamar, die auch die Grenze zu Cornwall bilden. Nach Osten bildete der River Plym die Stadtgrenze, durch Eingemeindungen erstreckt sich heute jedoch das Stadtgebiet auch östlich des Flusses. Etwa einen Kilometer südwestlich der Stadt liegt die zu Plymouth gehörige Insel Drakes Island im Plymouth Sound.
