Nantglyn heeft door de eeuwen heen verschillende opmerkelijke inwoners gekend, waaronder David Samwell (1751-1798), de scheepsarts aan boord van de Discovery tijdens de laatste ontdekkingsreis van kapitein Cook. Samwell hield een dagboek bij dat een gedetailleerd verslag van de reis bevat, en hij was getuige van Cooks dood door toedoen van vijandige inheemse bevolking op Hawaï in 1779. Dr. William Owen Pughe (1759-1835), een bekende literaire figuur die onder andere een Welsh-Engels woordenboek en een Welshe grammatica samenstelde, woonde de laatste tien jaar van zijn leven in het dorp, hoewel hij er niet geboren was.
