Basilika St. Jakob er en romersk-katolsk kirke i den gamle by i Prag, Tjekkiet. Den bærer titlen Basilica minor og er en del af verdensarven Historiske Center i Prag.
Den første kirke blev her bygget i gotisk stil sammen med etableringen af minoriterklosteret i 1232 af Wenzel I. Den præcise placering af denne oprindelige kirke og dens udseende er ikke kendt. I kirken fandt kroningsmåltidet for kong Albrecht af Böhmen og Elisabeth sted i 1311.
