Kilderne til Mainen udspringer i Fichtelgebirge og i Fränkische Alb. Ved byens vestlige kant Kulmbach i bydelen Melkendorf nær slottet Steinenhausen forenes de to kilder til selve Main. Flodens løb holder på trods af mange markante retningsovergange sin – i Central-Europa sjældne – hovedstrømning fra øst mod vest og berører flere franske middelhøje. Ved Main ligger store dele af det franske vinområde og adskillige, delvist velbevarede historiske bykerner. Store byområder gennemstrømmes af Main ved Würzburg og Frankfurt. Over for Mainz’ gamle bydel – mellem Ginsheim-Gustavsburg og Maaraue i Mainz-Kostheim – munder den ud i Rhinen. Fra dette punkt opstrøms til oven for jernbroen ved Hallstadt er Main en statslig vandvej.
