Penne-d'Agenais er en kommune med 2.341 indbyggere i Frankrig i departementet Lot-et-Garonne i regionen Nouvelle-Aquitaine. Beliggende på en klippefremspring dominerer bastiden Penne-d'Agenais dalen ved Lot med 120 meter. Ved dens fod ligger stedet Port de Penne, ved sammenløbet af Lot med den venstre biflod Boudouyssou, som optager sin biflod Tancanne cirka 1 km før. På den modsatte side, på højre bred af Lot, ligger kommunen Saint-Sylvestre-sur-Lot.
