Byen Cassino ligger i den italienske provins Frosinone, i regionen Lazio. Historisk set tilhører byen den mere sydligt beliggende region Campania. Det er et vigtigt kommercielt, landbrugs- og industrielt center.
Stedet var beboet længe før romernes ankomst af italienere. I den romerske periode hed byen Casinum. Fra denne tid stammer det velbevarede teater og amfiteater i Cassino. Ved byen ligger også en del af den oprindelige Via Latina, der forbandt Rom med Casilinum.
