Universitetet stammer fra Det Kongelige Akademi for uddannelse af civilingeniører både til statens tjeneste og til industrien samt af elever til handelsuddannelse, som blev grundlagt den 8. januar 1842 med støtte fra kong Willem II. Den første direktør og grundlægger var Antoine Lipkens. Kort efter stiftelsen fik akademiet sin første bolig på Oude Delft 95.
