Der Landkreis Peine erstreckt sich in etwa auf halber Strecke zwischen den beiden niedersächsischen Oberzentren Hannover und Braunschweig. Im Süden des Kreisgebiets hat er Anteil an den fruchtbaren Ackerböden der Hildesheimer Börde. Etwa nördlich der Bundesautobahn 2 machen sich im Norden des Kreises bereits Ausläufer der Südheide bemerkbar. Die Böden werden hier sandiger.
Der Landkreis liegt nördlich der Mittelgebirgsschwelle bereits in der Norddeutschen Tiefebene. Höchste natürliche Erhebung ist mit 132 m ü. NHN der Ballenberg, westlich der Ortschaft Barbecke in der Gemeinde Lengede, an der südlichen Kreisgrenze zum Landkreis Hildesheim und zur Stadt Salzgitter.
