Umgeben wird Mézilles von den Nachbargemeinden Tannerre-en-Puisaye im Norden, Dracy im Nordosten, Fontaines im Osten, Saint-Sauveur-en-Puisaye im Süden, Ronchères im Südwesten sowie Saint-Fargeau im Westen und Südwesten.
