Esta edificación fue habitada por tres generaciones de la familia Colón. Su construcción se inició a finales de 1510 y en el 1512 ya vivía allí Don Diego Colón, virrey de Las Indias, tercer gobernador de La Española e hijo del descubridor de América.
El Alcázar es una planta rectangular de dos niveles y fue construido con piedra coralina proveniente de las canteras de Santa Bárbara.
En la Actualidad es un museo relacionado al período virreinal en la República Dominicana.
