La città si trova sul margine nord-orientale dell'Harz. Il Selke attraversa il paese, in cui si trovano diversi stagni. Tra questi c'è anche il Pappelteich.
Ermsleben viene citata per la prima volta nel 1045 con il nome di "Anegremislebo" come proprietà dei conti di Falkenstein. Già nel 1298 il luogo era indicato come "oppidum". La chiesa, molto probabilmente più antica, di San Sisto, che successivamente subì numerosi cambiamenti strutturali, viene menzionata per la prima volta nel 1296. Il luogo ottenne il diritto di mercato nel 1530.
