La ville est située à l'extrémité nord-est des Harzes. La Selke traverse le village, où se trouvent plusieurs étangs, dont l'étang des peupliers.
Ermsleben a été mentionné pour la première fois en 1045 sous le nom de « Anegremislebo » en tant que propriété des comtes de Falkenstein. Déjà en 1298, le lieu était désigné comme « oppidum ». En 1296, l'église probablement plus ancienne de Saint-Sixte, qui a ensuite été plusieurs fois modifiée, est mentionnée pour la première fois. Le lieu a obtenu le droit de marché en 1530.
