Situato in posizione nodale per il medio-basso Salento, è un importante centro industriale ed economico della provincia. Comprende anche la piccola frazione di Morigino.
Il territorio comunale di Maglie, che si estende nella parte centrale della provincia, circa 29 km km a sud del capoluogo, con una superficie di 22,36 km², è situato nell'entroterra idruntino, nell'estremità meridionale della pianura salentina, un vasto e uniforme bassopiano del Salento compreso tra i rialti terrazzati delle Murge a nord e le Serre salentine a sud.
