Il centro sorge su una collina che domina da un lato la piana e la valle dell'Albegna e dall'altro lato l'area del Tufo.
Il territorio comunale di Manciano si estende nell'entroterra della Toscana meridionale, nel territorio delle colline dell'Albegna e del Fiora. L'estremità occidentale digrada nella pianura maremmana, lungo il corso del fiume Albegna, a valle della località di Marsiliana, mentre l'estremità nord-orientale penetra nell'area del Tufo lungo il corso del fiume Fiora che, da nord a sud, attraversa la parte orientale del territorio comunale.
