Groot Quassow is een deelgemeente van de gemeente Userin in de Landkreis Mecklenburgische Seenplatte in Mecklenburg-Voor-Pommeren.
De plaats ligt drie kilometer zuidoostelijk van Userin. De buurdorpen zijn Lindenberg in het noorden, Vorderster Kalkofen in het noordoosten, Voßwinkeler Schleuße in het oosten, Voßwinkel in het zuidoosten, Below in het zuiden, Klein Quassow in het zuidwesten, Useriner Mühle in het westen en Userin in het noordwesten.
