Der Palast wurde Anfang des 16. Jahrhunderts von der Familie Lanfranchi erbaut und erhielt Ende des Jahrhunderts nach Renovierungsarbeiten sein heutiges Aussehen: Ascheverputz, Ädikula und Kniefenster. Der Name Tuscanylli wurde 1827 geprägt, als der Palazzo von der gleichnamigen Familie gekauft wurde. Im Inneren sind die Decken mit Fresken von Nicola Cianfanelli, Gaspero Martellini und Annibale Gatti geschmückt, die Szenen darstellen, die Byron und Poesie, Galileos Die Apotheose und Michelangelos Die Apotheose darstellen (dem das Design des Gebäudes zugeschrieben worden sein soll).
