Habituellement nommée simplement Santo Spirito, elle est située dans le quartier Oltrarno. Elle est de style architecture Renaissance.
Elle fut construite sur les ruines d'un couvent augustin du XIIIe siècle, détruit par un incendie en 1371. Ce couvent était un important centre intellectuel, comprenant écoles, hospices, réfectoires pour les pauvres et une bibliothèque qui fut enrichie par le legs de Boccace.
