La cittadina, sede vescovile dal VI secolo, si trova lungo la costa occidentale del Salento, protesa sul mar Ionio. La città è divisa in due zone distinte: il centro storico, che sorge su di un'isola di natura calcarea e ha un circuito di circa 1,5 km, e il borgo nuovo, collegato all'isola mediante un ponte in muratura risalente al XVII secolo, in seguito alla qual costruzione si è formato progressivamente il borgo nuovo che si estende ad oggi su un'area superiore a quella di tutta l'isola. A ovest di Gallipoli sorgono l'isola del Campo, l'isolotto chiamato de "Li picciuni" e l'isola di Sant'Andrea che, completamente pianeggiante, si estende per circa 50 ettari.
