È la più estesa dell'arcipelago delle Pelagie nel mar Mediterraneo, nonché il territorio italiano più meridionale in assoluto, e fa parte del libero consorzio comunale di Agrigento, geograficamente si trova in Africa. Amministrativamente forma assieme a Linosa il comune di Lampedusa e Linosa , che conta 6 373 abitanti complessivi. Con una superficie di 20,2 km², è la quinta per estensione delle isole siciliane. In greco antico era nota come Λοπαδοῦσσα Lopadoûssa, poi latinizzato in Lopadusa. Appartiene alla placca africana.
