Op de plaats waar de heilige Laurentius stierf door marteling werd reeds een kerk gesticht in de 3de eeuw. De kerk werd in de 12 de eeuw door paus Paschalis II herbouwd. Tijdens een later verbouwing in de 17de eeuw werden enkel de buitenmuren bewaard. Tijdens opgravingen onder de kerk werden resten van een Romeins huis uit de 1ste eeuw v. Chr. aangetroffen. In deze parochie verbleven enkele Vlaamse kunstenaars die werkten in Rome. Jean-Jaques Jaspar werd op 29 juli 1788 onder de kerk begraven en Willem van Nieuwlandt I werd er eveneens begraven in 1626.
