Het paleis werd aan het begin van de 16e eeuw gebouwd door de familie Lanfranchi en kreeg zijn huidige uiterlijk aan het eind van de eeuw na verbouwingen: bepleistering met as, aedicula en knieschotvensters. De naam Toscanelli werd in 1827 bedacht toen het palazzo door de gelijknamige familie werd gekocht. Binnen zijn op de plafonds fresco's te zien van Nicola Cianfanelli, Gaspero Martellini en Annibale Gatti met scènes die Byron en poëzie uitbeelden, De Apotheose van Galileo en De Apotheose van Michelangelo (aan wie het ontwerp van het gebouw zou zijn toegeschreven).
