Dette fort blev bygget i 1584, midt i den Eighty Years' War. Opdraget kom fra feltmarskal Alexander Farnese. På anmodning fra den spanske konge forsøgte han at få de oprørske Nederlande tilbage under kontrol. Med en skibsbro blev fortet Sint-Marie forbundet med fortet Sint-Filips, på den anden side af Schelde. Således afskedede Farnese forsyningen til Antwerpen. Hans plan lykkedes: i 1585 måtte Antwerpen overgive sig.
