Dit fort is gebouwd in 1584, in volle Tachtigjarige Oorlog. De opdracht kwam van veldheer Alexander Farnese. In opdracht van de Spaanse koning probeerde hij de opstandige Nederlanden weer onder de knoet te krijgen. Met een scheepsbrug werd het fort Sint-Marie verbonden met het fort Sint-Filips, aan de overkant van de Schelde. Zo sloot Farnese de bevoorrading van Antwerpen af. Zijn plan slaagde: in 1585 moest Antwerpen zich overgeven.
