Burkau ligger i centrum af Bautzen-distriktet i et skovrigt område i Oberlausitz. Byen ligger i dalen ved den unge Klosterwasser, der udspringer i den vestlige ende af byen. Derudover ligger kilden til den Schwarze Elster i det nordvestlausitziske bakkeområde.
Kommunen ligger cirka 6 km nord for Bischofswerda og 16 km vest for kredsbyen Bautzen. Den ligger ved motorvejen A4. Tilslutningspunkterne Burkau og Uhyst am Taucher ligger i kommunens område.
Baneovergangen Bischofswerda–Kamenz blev nedbrudt, men stationsbygningen i Burkau er bevaret.
