Monistrol-sur-Loire er en kommune i det franske department Haute-Loire. Kommunen havde 8.875 indbyggere pr. 1. januar 2019. Stedet er en del af arrondissementet Yssingeaux. Omkring år 900 eksisterede der allerede en sogn i Monistrol. Denne fik overdraget relikvierne fra den hellige Marcellin. Omkring år 1000 blev der bygget en borg på en højde over kirken. I midten af det 12. århundrede blev der bygget en ny romansk kirke, hvis dele stadig kan ses i den nuværende kirke. Herren af Monistrol var fra 1270 biskoppen af Le Puy-en-Velay, som regelmæssigt boede i Monistrol i biskopslottet. Dette slot fra det 13. århundrede blev udvidet i det 15. og 17. århundrede.
