Under den meget varme (op til 42 °C) 13. juli 1967 faldt Simpson 1,4 km før toppen af bjerget af sin cykel. Hans sportsdirektør satte ham op igen, men Simpson faldt igen. Han blev bevidstløs og blev fløjet med helikopter til hospitalet i Avignon, men døde på vej dertil. Den daværende Tour-direktør Felix Lévitan blev kritiseret for at have fastlagt en for hård rute. Senere blev der dog fundet fire ubrugte ampuller i Simpsons skjorte, hvoraf en indeholdt amfetamin. Simpson døde sandsynligvis af en kombination af varme og udmattelse. Men det var ikke kun brugen af doping, der var årsagen til hans død. Det var kombinationen af ekstrem varme, dehydrering, hans vilje til aldrig at give op, alkohol og brugen af amfetamin, der førte til hans fatale skæbne. Simpsons død førte til strammere dopingkontroller under Tour de France.
