Otranto Domkirke er en romersk-katolsk domkirke i den italienske by Otranto, dedikeret til bebudelsen af Jomfru Maria. Det er ærkebiskoppens sæde for ærkebispedømmet Otranto. Domkirken blev indviet i 1088. Den er 54 meter lang og 25 meter bred og er bygget på 42 monolitiske granit- og marmor søjler fra ukendte brud. Dens plan er en tre-naves kirkerum med en apsidal østende. På hver side af den vestlige facade er der to lancetvinduer.
