Vila Real de Santo António er en by i den portugisiske kommune Vila Real de Santo António og har 11.946 indbyggere. Det er den sydøstligst beliggende by i provinsen Algarve og landet Portugal. På den østlige side af byen løber floden Guadiana, som også danner grænsen til Spanien. Det er muligt at komme fra Vila Real de Santo António til den anden side af floden både med bil og med båd. På den sydlige side af byen ligger en sandstrand ved Atlanterhavet.
