Der Herzog von Brabant ließ zu Beginn des 12. Jahrhunderts ein Gebetshaus an dem Weg errichten, der Brüssel von Osten nach Westen durchkreuzte. Es ist möglich, dass dieses Gebetshaus ursprünglich die Hofkapelle des ersten Schlosses war, das um 1100 von den Grafen von Löwen auf dem Koudenberg, dem höchsten Hügel des linken Ufers der Zenne, errichtet wurde. Es ist auch möglich, dass die St.-Jakobskirche mit einem Hospital für Pilger verbunden war, das in der Nähe des zuvor genannten Schlosses lag. Dies würde erklären, warum St. Jakob als Schutzpatron gewählt wurde. Die Existenz der Kirche wurde bereits im 12. Jahrhundert beschrieben; die Namen der Bediensteten sind ab dem Jahr 1121 bekannt.
