Markranstädt befindet sich in der Leipziger Tieflandsbucht etwa zehn Kilometer südwestlich vom Leipziger Zentrum. Die Stadt grenzt im Norden an Schkeuditz, im Osten an Leipzig, im Süden an Pegau sowie im Westen an die in Sachsen-Anhalt liegenden Städte Lützen, Bad Dürrenberg und Leuna. Sie liegt am Westufer des Kulkwitzer Sees.
Markranstädt besteht neben dem eigentlichen Stadtgebiet aus den 17 Ortsteilen in sechs Ortschaften:
