Die Stadt an der Großen Röder liegt etwa 20 Kilometer östlich der Landeshauptstadt Dresden. Sie zieht sich südlich von Pulsnitz etwa vier Kilometer an der Alten Poststraße entlang. Autobahnanschlussstellen sind Ohorn und Pulsnitz an der A 4.
Die nächstgelegenen Ortschaften im Umkreis der Stadt sind die unmittelbar östlich anschließenden Großröhrsdorfer Ortsteile Bretnig und Hauswalde sowie weiter östlich Rammenau, weiterhin der Ortsteil Kleinröhrsdorf, die nicht zu Großröhrsdorf gehörenden Ortsteile Leppersdorf, Lichtenberg, Wallroda und Seeligstadt, die Gemeinde Ohorn sowie die Städte Radeberg und Pulsnitz.
