Das Herrenhaus Schloss Melkof liegt an einem See im Vellahner Ortsteil Melkof, einer Gemeinde im Landkreis Ludwigslust-Parchim in Mecklenburg-Vorpommern. Es befindet sich heute in Privatbesitz. Das Gebäude ist mit seinen über 4800 m² Nutzfläche eines der größten Herrenhäuser Norddeutschlands aus der Zeit des Historismus. Gebäude und Park liegen im Zentrum des Mecklenburgischen Elbauetals.
In einer Urkunde vom 27. August 1471 wurde Knappe Ullrich Pentz zu Melkhof erstmals urkundlich erwähnt. Aus dieser ist zu entnehmen, dass er Eigentümer von Hufen und Höfen in Melkhof war. Ullrich Pentz gilt als Stammvater des Hauses Melkhof.
