Das Amt Demmin-Land hat seinen Sitz in der amtsfreien Stadt Demmin. Zur Erledigung ihrer Verwaltung sind 16 Gemeinden zusammengeschlossen. Das Gebiet des Amtes erstreckt sich im Norden des im Zuge der Kreisgebietsreform 2011 neu geschaffenen Landkreises Mecklenburgische Seenplatte im Land Mecklenburg-Vorpommern. Historisch gesehen gehört Demmin und das sogenannte Demminer Land aber zu Vorpommern.
Dem Amt, das seit 1994 existiert, gehörten ursprünglich elf Gemeinden an. Am 1. Juni 2004 wurden fünf vormals selbständige Orte eingemeindet: Hohenbrünzow , Quitzerow , Sanzkow , Teusin , Wotenick . Am 1. Juli 2004 kamen auch die zehn Gemeinden des aufgelösten Amtes Borrentin zum Amt Demmin-Land hinzu.
