Von Emden aus folgt die zweite Etappe dem Larrelter Tief in Richtung Mahlbusen. Dann geht es weiter durch die kleinen Ortschaften Rysum, Loquard und Campen in Richtung Nordseeküste. Nach einem kurzen Abschnitt hinter dem Deich biegt die Route wieder etwas landeinwärts ab in Richtung Grothusen, Manslagt und Neu Etum.

Bei Routenkilometer 41 wird Pilsum erreicht. Das Runddorf wurde auf einer Warft (Siedlungshügel) errichtet. Besonders bekannt ist der Pilsumer Leuchtturm. Der rot-gelb geringelte Leuchtturm ist eines der Wahrzeichen Ostfrieslands und diente bereits des Öfteren als Filmkulisse. Vom Leuchtturm aus sind es nur noch wenige Routenkilometer bis zum Etappenende, dem Fischer- und Künstlerdorf Greetsiel.
