Die Einwohner heißen Ménesplesiens.Ménesplet liegt im Périgord am Fluss Isle, in den hier der Duche mündet. Die Nachbargemeinden von Ménesplet sind Montpon-Ménestérol im Norden und Osten, Saint-Martin-de-Gurson im Süden, Moulin-Neuf im Westen sowie Le Pizou im Nordwesten.
