Tarsus liegt etwa 30 km nordöstlich der Provinzhauptstadt Mersin, am Treffpunkt der Autobahn Mersin-Adana mit der aus Norden durch die Kilikische Pforte kommenden Europastraße 90. Seit einer Gemeindereform 2014 ist die Kreisstadt flächen- und einwohnermäßig identisch mit dem Landkreis.
Die Hafenstadt Tarsus am Golf von İskenderun, die Handelsbeziehungen nach Phönizien und Ägypten unterhielt, lag ca. zwei bis drei Kilometer vom Mittelmeer entfernt und war über den schiffbaren Fluss Kydnos erreichbar. Der Hafen ist heute verlandet, und die Stadt liegt etwa 16 km vom Meer entfernt.
