Le comté de DeSoto est un comté des États-Unis situé dans l'État de Floride, aux États-Unis. Sa population était de 34 862 habitants en 2010. Son chef-lieu est Arcadia. Le comté a été fondé en 1887 et doit son nom à Hernando de Soto, un explorateur espagnol. Jusqu'en 1921 le comté était plus grand et comprenait les comtés de Charlotte, de Glades, de Hardee et de Highlands.
Selon l'American Community Survey, en 2010 71,53 % de la population âgée de plus de 5 ans déclare parler l'anglais à la maison, 26,66 % déclare parler l'espagnol et 1,81 % une autre langue.
