Het originele kasteel werd in de vroege middeleeuwen gebouwd door de Agilolfingen. Dit werd in 1247 verworven door de hertogen van Wittelsbach.
Toen graaf palatijn Otto Hendrik in 1522 zijn heerschappij begon in Palts-Neuburg, vond hij een middeleeuws versterkt kasteel in zijn woonstad Neuburg, dat, in tegenstelling tot vergelijkbare prinselijke residenties, nog niet was aangepast aan de eisen van een moderne hof. Daarom gaf hij vanaf 1527 opdracht om het kasteel opnieuw te ontwerpen tot een renaissancepaleis en om de artistieke kwaliteit en staat uit te breiden tot een van de belangrijkste paleizen van de eerste helft van de 16e eeuw in Duitsland. Vanaf 1537 werd er een extra westvleugel toegevoegd, die ook de kapel omvat.
