Het paleis werd in opdracht van Domenico della Rovere, kardinaalpriester van San Clemente en neef van paus Sixtus IV, gebouwd tussen 1480 en 1490 door de architect Baccio Pontelli in het Borgo. De Florentijnse architect Pontelli hield zich nauw aan de architectonische stijl van het Palazzo Venezia, een van de belangrijkste gebouwen in de 15e eeuw in Rome. Meer dan 300 jaar lang stond het paleis bekend als het Palazzo dei Penitenzieri. Het paleis staat onder monumentenzorg en is alleen toegankelijk in het hotelgedeelte.
