Holwerd ligt ten noordwesten van Dokkum en ten zuiden van de Waddenzee, tussen Blija, Waaxens en Ternaard. De dorpskern ligt ingeklemd tussen de N356, N357 en N358. De Holwerdervaart stroomt langs de N356 naar het zuiden toe, en heeft tevens een kleine aanleghaven.Tussen het buitendijkse gebied en het dorp is een klein industrieterrein. In 2021 telde het dorp 1.595 inwoners. Onder het dorp vallen ook de buurtschappen Kletterbuurt en Drieboerehuizen.
