Het ligt nabij de Waddenzee, ten noordoosten van Franeker en ten noordwesten van Leeuwarden, tussen Sint Jacobiparochie en Vrouwenparochie. Door het dorp stromen de Noordervaart en de Zuidervaart, en verder onder meer de Blikvaart en de Zuidhoekstervaart. De N383 en de N393 komen samen in het dorp.In 2021 telde Sint Annaparochie 4.830 inwoners. Het is daarmee het grootste dorp van de gemeente Waadhoeke, alleen de stad Franeker heeft een groter inwonertal. Tot de gemeentelijke herindeling op 1 januari 2018 was Sint Annaparochie de hoofdplaats van de voormalige gemeente het Bildt.
