Den Store Lienewitzsø er en sø i Landkreis Potsdam-Mittelmark i delstaten Brandenburg. Den er cirka 13,78 hektar stor og strækker sig cirka 740 meter fra nordøst til sydvest. Den bredeste del er cirka 200 meter med en maksimal dybde på 6 m. Den ligger i området for den amtsfrie kommune Michendorf.
Den Store Lienewitzsø ligger syd for Havel i et skovområde i naturbeskyttelsesområdet Lienewitz-Caputher Søers og Vådeområdernes kæde. I nord har den en grøftlignende forbindelse til Caputher Sø. På grund af det faldne grundvandsniveau er denne forbindelse for det meste tør. I den sydvestlige ende ligger beboelsen Lienewitz. Sydvest for vandet ligger den Lille Lienewitzsø. Motorvejen A10, den sydlige Berliner Ring, ligger cirka en kilometer væk.
