Karlsburg ist eine Gemeinde im Landkreis Vorpommern-Greifswald. Die Gemeinde wird vom Amt Züssow mit Sitz in Züssow verwaltet. Die Gemeinde liegt südlich der Kreisstadt Greifswald. Mit Wirkung zum 26. Mai 2019 ging die bisherige gleichnamige Gemeinde Karlsburg in der gemeinsam mit Lühmannsdorf neugebildeten Gemeinde Karlsburg auf.
Karlsburg liegt 21 Kilometer südöstlich der Kreisstadt Greifswald und 14,5 Kilometer östlich von Gützkow, nach Wolgast dem Zugang zur Insel Usedom sind es 19 Kilometer. Im Karlsburger Holz ist die Quelle der Swinow, die die ganze Gemeinde quert. Im Ortsteil Steinfurth beginnt der Brebowbach. Stillgewässer sind nur in Form der Sölle vorhanden.
