Tussenhausen liegt circa 35 Kilometer östlich von Memmingen in der Region Donau-Iller in Mittelschwaben.
Kaiser Friedrich III. erhob den Ort im Jahr 1455 zum Markt. Tussenhausen gehörte der Herrschaft Angelberg bzw. Herrschaft Schwabegg und wurde um 1680 Teil des Kurfürstentums Bayern. Tussenhausen besaß seit 1455 das Marktrecht mit wichtigen Eigenrechten. Im Zuge der Verwaltungsreformen im Königreich Bayern entstand mit dem Gemeindeedikt von 1818 die heutige Gemeinde.
